
Sirius Red Staining Solution
Introduction
Sirius Red Staining: Collagen is basic in nature and reacts strongly with acidic dyes. Sirius Red is a highly acidic dye that binds to collagen fibers, producing a distinct birefringence effect. With Sirius Red staining, collagen fibers appear red, cell nuclei appear green, and other tissue components appear yellow.
Collagen fibers are the most widely distributed and abundant type of fiber in connective tissue and are present in a variety of organs. Type I collagen fibers are mainly found in bone, skin, and tendons; Type II collagen fibers are primarily present in cartilage; Type III collagen fibers are mainly distributed in embryonic tissues, adult blood vessels, and the gastrointestinal tract; and Type IV collagen fibers are mainly found in basement membranes.
