
Endospore Staining Solution
Introduction
Endospores are highly resistant dormant structures formed by bacteria of the genera Bacillus and Clostridium at a certain stage of growth. They are typically round or oval in shape. Endospores have thick walls and low permeability, making them difficult to stain and decolorize.
Therefore, when a strongly staining dye is applied under heated conditions, the dye can penetrate both the vegetative cells and the endospores. The dye within the vegetative cells can be removed by washing with water, whereas the dye that enters the endospores is difficult to wash out. After counterstaining, the endospores retain the color of the primary stain, while the vegetative cells take on the color of the counterstain. As a result, the vegetative cells and endospores are stained red and green, respectively, making them easy to distinguish.
