Introduction

The capsule is a distinctive mucous-like substance formed outside the bacterial cell wall during bacterial growth and survival. Bacterial capsules can protect bacteria against phagocytosis and digestion, thereby increasing their invasiveness.


Because capsules have low light-blocking properties and poor affinity for dyes, they are not easily stained by conventional staining methods. Special staining techniques are therefore required for their observation. Methods for demonstrating bacterial capsules include the Hiss method, Muir method, Anthony method, Tyler staining method, and India ink staining method. The Muir method involves relatively complex reagents and procedures and is therefore rarely used at present. The most commonly used methods are Tyler staining and India ink negative staining.
